in-situ顕微鏡法とは?
In-situ透過型電子顕微鏡法では、透過型電子顕微鏡(TEM)の像観察機能をさまざまな外部刺激の作用と組み合わせてリアルタイムで結果を観察、計測します。現在では、多種多様なシステムおよびホルダーを利用して、電気的なバイアシング、機械的なひずみや変形、加熱、冷却、ガスや液体環境の導入、そして単に強い電子線の照射といったさまざまな刺激を与え、動的な反応を評価することが出来ます。
過去において、さまざまな反応や系の変化の映像はビデオテープに記録されてきました。その後、スクリーンキャプチャープログラムを用いることで、発生している現象を低解像度の定性的な毎秒30フレーム(fps)ほどのビデオとして取得するまでには改善されました。
高速データ転送機能と高速データ処理機能が実現されたことで、GatanのISカメラではセンサーからの出力を巨大なデータセットとして直接記録し管理できるようになりました。実験中の各取得フレームからのオリジナルの定量的なデータが直接ストレージへと保管され、それぞれを独立した像として取り扱ったりビデオとして再生することができます。加えて、さまざまなアルゴリズムやスクリプト(画像積算、ドリフト補正、ビニングなど)を取得後に適用して、ビデオで観察された取得データ中の最も有用な部分を抽出可能です。GatanのISカメラに備えられた組み込みデータ管理ツールを使用することで簡単にデータを切り取って抽出することが出来、系に加えられた刺激の効果を定量化するためのデータの後処理が実現されます。このデータ取得と後処理のワークフローによって、電子顕微鏡観察中の測定回数を大幅に増加させることが可能です。これまで実現不可能であったサブミリ秒の時間分解能と空間分解能での測定を達成し、データ取得には速すぎるために捉えることが出来なかった反応の研究を可能とし、これまで未知であった反応過程の解明を実現しました。
本データは、1300℃でのGatan社製OneView® IS カメラとDENSsolutions社製Wildfire加熱試料ホルダーの双方の安定性を示しており、優れた空間分解能と時間分解能で取得した温度変更直後のナノ粒子の形状変化を捉えています。いずれの粒子が変形するかは判りませんが、 OneView IS カメラの広視野によって同時に等しい空間分解能で全ての粒子を観察することが出来ます。
本ビデオは10倍速で再生していますが、粒子の形状変化は2倍速で再生しています。挿入図は拡大表示しており、格子が解像されていることを示しています。

SiNx上のAuスパッタ粒子の焼結過程を、Gatanの Rio™ 16 IS カメラで取得しました。DENSsolutions社製Wildfire加熱試料ホルダーを使用して、試料を650℃まで加熱しています。本ビデオはGatan Microscopy Suite® ソフトウェアのIS player機能を使用してドリフト補正と切り出し作業を行っています。ビデオの高解像度版をダウンロードされたい方はこちらをクリックしてください。
本データは微細なCeO2粒子の表面における個々の原子コラムの移動を400fpsで捉えていることを示しています。この動きは室温での電子線照射によって引き起こされています。 K2® IS カメラの時間分解能によって、これらの動きを捉えることが可能になりました。
本ビデオは二つの異なる速度(実速度の1/2と1/10)で再生しています。そのため、動画中の変化を詳細に捉えることが可能です。1/10での再生速度においても、まだいくつかのフレームはまだスキップしています。これは1/10での再生速度でもまだ40フレーム/秒であり、それに対してビデオフレームレートは30フレーム/秒であるためです。
